Your John Plasters is not a son of Isaac Plasters. The best candidate appears to be Thomas Plasters, Jr. and Elizabeth Batts.(see History/Biogs. of Cass Co.) Isaac and Thomas, Jr. had a brother, Lemon Plasters, but he didn't marry until 1840, so he couldn't be the father of John. In the 1840 census, Thomas, Jr. has a son in the age category 0-5, which would be John's age at that time. (b. 1838-39).
Your best bet would be to check the Cass Co. website for any records online that would help you. Also, probate records listing heirs of Thomas, Jr. would confirm John's belonging to that family. Try this website:

Why John is in another household in 1850 may be a clue to follow? Also, look under both spellings, Plaster or Plasters. I have found it under Plaister in some records.
