There is a James boarding with a family in West Township on the 1910 Census that could be your man. Also a married couple with the Singleton name that is probably related.

James Singleton, 23yo, Single, MO MO MO
Oscar Singleton, 21yo, M1yr, MO MO MO, Farmer
Lula Singleton, 21yo, M1yr, MO MO MO

There is also an Arthur Singleton and a J.T. Singleton that could be related.

Head of family is Dempsy Gardner
Arthur Singleton, 17yo, Single, Hired Man, MO MO MO, Farm Hand

1910 Missouri, new Madrid County, Big Prairie, ED#106, Page 80B:
J.T. Singleton, head, 24yo, M3yrs, MO UNK UNK, Laborer
A.M. Singleton, wife, 23yo, 2 children/2 living, MO UNK UNK
J.E. Singleton, son, 3yo, MO MO MO
A.N. Singleton daughter, 7/12, MO MO MO

There are no Singletons living in New Madrid County in 1900. Good luck!
