There are a couple of MILLS families who were in the area of Elgin ON CAN in the time frame of which you are in search.
Before and after.

Have you looked at those families found on www.rootsweb.ancestry.com and www.gencircles.com as well as Ancestry.com???

They may tie into your John BURDEN ? MILLS and Jane.

As well there is a John B MILLS, B: 1839 son of Cornelius MILLS and Nacy BARRY, but he is of Markham twp, York, ON CAN
and apparently removed from there to Pike Co. Illinois USA, where he lived on and died.

I believe some DNA testing has been done on both of these particular MILLS families. See www.familytreedna.com under MILLS go to DNA results page, haplo group R and I (eye).
