You may indeed be right that there was another wife by the name of Mattingly. I do not have record of her but William Coomes who married Elizabeth Peyton did have another wife I do know of named Martha Hite. Given that, it is not inconceivable that under extraordinary circumstances (being widowed, divorced, etc.) that there could have been a third wife.
The Coomes family intermarries into the Alvey family and there was an earlier intermarriage between the Alvey's and a family named Clarke. One generation from that marriage is the marriage of a Clarke to a Mattingly so there does appear to be a geographic and historical connection between these families.
All I can say is that all documented records for my family line have William Peter's spouse as Elizabeth Peyton. (That is to say, the mother of Susan Isabella Coomes who married William Henry Alvey was Elizabeth Peyton.)
Sorry I can't give you any further information on the Mattingly/Coomes marriage of which you wrote but remember, it takes all of us to build a tree.
