I searched the Irish Family History Foundation's database. It contains 12,000,000 surviving rcds for abt 65% of Irish counties. Rcds are sparse B4 1840, rare B4 1800. My search is limited to the 65% of Irish counties.
There are 33 variants of the Manning surname.
For the existing database, here is the distribution of the Manning surname based on the # of births 1700-1920:

For Michael Manning B 1848 (exact), I found 2 B rcds: Meath and Kilkenny. The Meath rcd had a father named John: the Kilkenny rcd Patrick. When you combine this fact with the brother's names, Meath appears to be the most likely.
For Hannah McGuillicuddy (one of many variants), I found 1 B rcd in the current database: 1874 Cork. This may not fit your timeline. Her rcds may be undigitized. She may have been born in the U.S. Chk the 1880 and 1890 censuses.
