There were two John Doty's in Johnson Co at that time and very distantly related. I am descended from the other one but I do maintain a small database on your line just so I can keep them straight.
I have Harriet born about 1808 and married to James Karr April 10, 1825, in Franklin Co IN. I have no descendants from them.
I have Harriets parents as John Doty (Born abt 1775 no location) and Catherine with no known last name.
I have John's parents as Benjamin and Mary.
I have that Benjamin's parents as Benjamin and Rachel.
I have that Benjamin's parents as Benjamin Doty and Abigail Whitehead.
I have this last Benjamin's parents as Samuel Doty and Jane Harmon.
I have Samuel's parents as Edward Doty (Mayflower) and Faith Clark.
However this is an unproven line from the Mayflower due to the fact there were several Benjamin's in NJ at the same time and close to same age. Maybe DNA will prove it one day!
I have seen the name Sarapheen connected to the Doty family but I am not sure exactly how she connects/
