According to what I have, John and Edward were the children of John, Sr's first marriage to Sarah Watkins and Frances "Fanny" was the daughter of the second wife, Mary Bass.

Mary Bass had been married before and had a son Jeremiah. She married for the third time 5 Sep 1771, Chesterfield Co., VA to Capt. Thomas Wooldridge.

John, Jr. and Edward are mentioned in their grandfather's [Edward Watkins] will in 1771. Fanny is not mentioned although she was born in 1759.
