An interesting question - both a moral one and a practical one.
Would it work in practice? What are the chances of extracting a useful yDNA sample? Could you do autosomal DNA as well. Perhaps the experts can tell us?
On the moral issue, I would say it depends on the feelings of the family alive today. If they are happy, go ahead. There is no way you can ask the person who is buried after all.
