The surname seems to have had its origins in the mountains of Santander, near Arnuero, at a site called Castillo. It was there that Alfonso de Venero Hernandez took the surname and became Alfonso del Castillo Hernandes. He was the great-grandson of Sancho Alfonso de Venero, who lived during the time of King Bermudo I, and King Alfonso II. All of Alfonso del Castillo's sons had the Castillo surname.

In the United States, Castillo is the 26th most popular surname among modern Hispanic families. In California in 1844, Maria Raymunda Castillo - daughter of Jose Castillo, of Mexico city, and Serafina Sinoba of Los Angeles - married Mariano de Jesus Soberanes at the San Antonio Mission. These children, Maria Concepcion, born 1831; Estefana del Rosario, born 1834; Maria Francisca, born 1836; and Maria Antonia, born in 1841, took the Castillo surname and were all were born in Monterey.

In Arizona, Juan Castillo lived in Tucson in 1820. Also in Tucson, but in 1831, were Carmen Castillo and her husband, Jose Lizarraga. Paulino Castillo's family, including Inocencio, Jose and Tomas, were in Tubac. In Santa Cruz, there were Joaquin and Juan Castillo, both in military families.

Mexico City in 1753 boasted more than 100 people with the Castillo surname. Among these were Antonio Castillo, age 40, a carpenter and widower, his son, Jose, 11, and daughter, Juana, 20. Cayetano del Castillo was a candle maker. Felipa Maria Castillo, a widow, lived with her daughter and they made chocolate for a living.

Don Juan del Castillo Velasco y Castro, born in Madrid, was Contador de La Real Armada, Secretario y Veedor de las Caballerizas de Felipe IV, y Alcalde de la Hermanidad por el estado de hijodalgo de Madrid. He was the head of the Castillo Velasco famlly that established itself in Peru and Chile in the 16th century.
