For the DNA testing you must have a direct male Brower descendant. And the best test to take would be the 37 marker test. As of now, no direct male descendants of Abraham Brouwer (wife Mary Elsworth) has participated in the test. However, if you did take the test, it would at least confirm (or refute) your ancestry from Adam Brouwer. Enough descendants of Adam Brouwer have tested to establish a DNA identity for Adam, so that at this point, others who are not sure, or who want to confirm their descent from Adam can easily do so.
Unless there was an adoption event in your family there is really no need for you, your father, or his brother to all be tested. The three of you should have identical test results. Anyone of the three of you would work for your purposes. Females don't have a Y chromosome so testing them would not tell you anything.
The Jane Stevenson person would be interesting to pursue. Finding her origins might yield some more info on Henry Brower (1806-1895).
