I'm afraid I can't add very much to what I've already told you. As I said, Mary was unmarried and I wasn't aware of any family. She came to my wedding in 1976 and if you ever establish a connection, I could send you a copy of a small photograph I have of a large group. It won't be very clear, but you might just be able to make out Mary's features and perhaps decide from that whether she was a descendant of Jeremiah.
Are you based in the UK or the US? Presumably you've looked at all the Nottinghamshire Brandreths listed in the 1841 census and tried to map out their relationships? One oddity I thought might be worth investigating is the 1 year old Timothy Brandreth living in Mansfield in 1841 with a family of a different surname. Given that people in those days often named their children after family members, he might be connected to Timothy who emigrated.
I don't know whether there would be any records from Ransome Hospital where Mary worked, listing her next of kin or anything. It no longer exists and my mother is also dead so I can't ask her. I'll ask my father next time I speak to him if he can remember anything, although Mary was my mother's friend so he may not be able to add much. I'll also ask my sister, whose godmother she was, but she's away on holiday at the moment so it'll be a while before I can get back to you.
