General Question, DNA Testing re Indian Heritage

Would DNA testing reveal whether or not there is American Indian ancestry, and if so, would the gender of the person being tested matter?

Family story has it that on my maternal grandmother's side, there is American Indian ancestry. Her parents were John Alfred Hall b. abt 1820 and Sarah Ellen ?Vermillion b. abt 1840. These families were from the Prince Georges and southern Anne Arundel County areas in Maryland. And I have no idea which branch the supposed Indian ancestry is from.

DNA? Yes? No? Appreciate any input.
